I have to disagree with the layer 5 bit, a good tileset is one that you dont NEED to use layer 5 for extra layer 4 stuff. Put it all in layer 4, so you don't need to use layer 5. Sure there are almost limitless combinations of tiles needed if you do this (Raging Inferno anyone?), but atleast make it very layer 4 friendly...
