You don't need to run your system on 256-colour mode to create tilesets. What you do need to do, however, is to reduce your tileset image to 256 colours. I'd suggest TilesetPal, an excellent tool for this purpose.

Well, creating the image is just the first step. Sometimes colour reduction can be a huge pain as well. Not to mention other considerations when creating textured backgrounds etc. Then there's also the mask, which goes along with your image. Ordinarily I'd redirect you to Disguise's Tileset Resource, but I have no idea where that is located anymore.
